Arvoituksellinen viesti menneisyydestä saa James Bondin (Daniel Craig) lähtemään yksin Mexico Cityyn ja sieltä Roomaan, missä hän tapaa pahamaineisen rikollisen kauniin ja kielletyn lesken, Lucia Sciarran (Monica Bellucci). Bond soluttautuu salaiseen kokoukseen ja paljastaa SPECTRE:nä tunnetun uhkaavan järjestön olemassaolon.

Sillä välin Lontoossa Max Denbigh (Andrew Scott) , Kansallisen turvallisuuden keskuksen uusi johtaja, kyseenalaistaa Bondin toimet ja myös M:n (Ralph Fiennes) johtaman MI6:n merkityksen. Bond värvää salaa Moneypennyn (Naomie Harris) ja Q:n (Ben Whishaw) avukseen etsimään käsiinsä Madeleine Swannin (Léa Seydoux), vanhan vihollisensa Mr Whiten (Jesper Christensen) tyttären, joka voi ehkä auttaa SPECTRE:n verkoston selvittämisessä. Koska Madeleine on salamurhaajan tytär, hän ymmärtää Bondia paremmin kuin kukaan muu pystyisi.

Uskaltautuessaan kohti SPECTRE:n ydintä Bondille selviää kylmäävä yhteys itsensä ja etsimänsä vihollisen (Christoph Waltz) välillä.
